Guaiacum officinale L. (Zygophyllaceae) is a medicinal and ornamental plant. It is commonly cultivated in the gardens in Pakistan. The present thesis describes the isolation and structure elucidation of six new tritrpenoid saponins namely guaianin M, guaianin N, guaianin O, guaianin P, guaianin Q, guaianin R as well as known saponin guaianin S (akebia saponin PE) form Guaiacum officinale L. Guaianins M, N, O and S were isolated fro the flowers while guaianin P, Q, and R were isolation for the bark of the plant. Both the flowers and bark were extracted in methanol and evaporated to gum separately. The gummy residues were partitioned between water and ethyl acetate and then water and butanol butanoilc extract contained saponins. Sapoinns were purified by VLC column chromatography, flash chromatography, and finally by HPLC through semi preparative RP-18 column. The structures of the saponins were elucidated with the help of chemical as well as spectroscopical analysis of the compound. The later included extensive studies of various techniques of both 1-D and 2-D NMR spectroscopes. From flowers we isolated a number of reported compounds. Besides them only three were new and one was reported for the first time form this plant. From the bark we isolated a number of reported compounds besides three new compounds guaianin P, Q and R. The chemical names and structures of guaianin M, N, O, P, Q,R and S are given below.
