Abstract
The work embodied in this thesis comprises two parts. The first part deals with the isolation of nine isoquinoline type alkaloids from Berberis waziristanica which has not hitherto been investigated. Identification of the eight known alkaloids was carried out by comparing their spectral data with those already reported in the literature. The unknown Waziristanine was also identified spectroscopically, mainly by 1H-NMR and mass spectra. Comparison of its spectral data with closely related alkaloid porveniramine proved more helpful in the identification of Waziristanine. Besides the usual spectroscopic techniques (UV, IR, 1H-NMR, MASS) we have also made use of the recently developed techniques including 2D-NMR, (COSY-45°, NOESY and 2D-J-resolved in order to give further support to the already established structures of the alkaloids.

Optical rotation or NOE measurements were rarely used to determine the relative stereochemistry of the asymmetric centres, BB and DEPT, experiments were carried out to determine the multiplicities of the carbon atoms.

The second part of the thesis is concerned with the determination by circular dichroism of the absolute stereochemistry of the cottonogenic derivatives prepared in situ from the chiral ligands (carboxylic acids, amino acids, glycols, amino alcohols nucleosides, nucleotides and amines) and chiral metal complexes of the general formula, (M3O(OOCR)6L3)כֿ+ where M = FE, CR, Mn, Rh, Ru, etc and L = water or pyridine.
