Abstract

In view of the problems such as nausea, vomiting, and gastric associated with oral administration of drugs, for which parenteral administration in some cases is not recommended, We made an attempt to prepare suppositories for some of the drugs for which such means drug delivery are not available. The suppositories of Diclofenac sodium, Ibuprofen, Amodiaquine hydrochloride and Chloroquine phosphate were prepared by fusion method with various bases including Cocoa butter, Witepsol H 15, Witepsol W 25, Witepsol W 35, and Witepsol E 75 and by non-fusion method in a mixture of Eudragit L & propylene glycol. Release rate of the drugs from these suppositories was dependent on the nature of base employed and quantitatively release was in the order Cocoa butter> Witepsol H 15 > Witepsol W 25 > Witepsol W 35 > Witepsol E 75. Formulation of Eudragit L suppositories indicated slow and insufficient release. The hydrophobic nature of lbuprofen presented slower release as compared to other drugs in the present study. Drug release from our formulations in the present work reveals a suitability of such preparations for therapeutic purpose, keeping in view the properties such as melting point, partition coefficient and dissolution in appropriate media. The bioavailability of the drugs from these preparations in rabbits corresponded well with the in vitro release of drugs from the suppositories.
