Abstract
Trichoderma harzianum was used as a biological control against Root-knot nametode (RKN) Meloidogyne javanica. Fifteen isolates of this fungus were isolated from soil, eggs, and eggs masses of Meloidogyne sp.,

Collected from the infested fields of Dargai and Swat Pakistan. The isolates were tested for their antagonistic activity against RKN. Culture filtrates (CF) were obtained from T.harzianum grown on potato dextrose broth.

Concentrations of the CF(25%,50%,75%,and 100%) were prepared. the highest egg hatching inhabitation (57.87%) and J2 mortality (66.73%) were recorded in T,harzianum isolate Th-1,collected from jabban. The concentrations of Trichoderma culture filtrates hade significant effect on egg hatching inhibitions and J2 mortality. The maximum egg hatching inhibition (90.16%) and the highest J2 mortality (92.57%) were noted in 100% CF concentrations. Jabbn isolate (th-1)was found more effective in parasitizing M.javanica eggs and J2 with the highesr egg parasitism (50.50%) and J2 parasitism (63.00%).On the basis of in vitro bioassays , four isolates of T.harzianum viz. Th-1,Th-2,Th-9,and Th-15 were selected.Genetic diversity among the fifteen isolate s was evaluated by using randomly amplified polymerfic DNA (RAPD). Four RAPD primers D-17,G-20,D-08 and H-02 were used to estimated the genetic distance among the isolates in all possible combinations, which resulted in high degree of genetic variability at molecular level among the various isolated of T.harzianum. The study revealed different levels of polymorphism for RAPD primers that result in amplification of 2.06,2.20,3.27 and 1.33 scorable bands (loci)per isolate of T.harzanum. A high degree of genetic polymorphism at DNA level was observed among the isolate with average genetic distance ranging 04.90%.T.harzianum isolates were classified into three distinct group. Most of the isolates had similar traits and were classified into group €“I. Group-II comprised of isolates Th-13 and Th15.while Group-III consisted of Th-4 ,Th-7 and Th-11. To study the efficacy of the selected trichoderma isolates, field trails were conducted in the Root-knot nematode-infested areas of dargai and swat, Pakistan The T.harzianum isolates ,grown on wheat grains substrate, were applied @ 8 g plant-1 either alone or in different combinations. It was observed that T.harizanum isolates significantly reduced the nematode populations. The fungus enhanced plant growth and yield in all the treated plots. Jabban isolates (Th-1) was found as the most effective in nematode suppression followed by Shamozai (Th-9) isolate.
