Abstract  

1. The bacteria isolated from different regions and organs of Labeo rohita (H) belonged to Actinobacillus, Alcaligenes, Corynebacterium, Enterobacter, Escherichia, Micrococcus, Pasteurella, Proteus, Pseudomonas, Serratia, Shigella and Staphylococcus genera. 2 Aeromonas hydrophila was isolated from the kidney and abscess present in the head of fingerlings of Labeo rohita suffering from tail rot, fin rot and haemorrage spots. 3. a) Intraperitoneal injection of live Pseudomonas fluorescence into the fingerlings of L. rohita at a concentration of 5x109, 5x107 and 5x10S cells/g body weight, resulted in 76%, 62% and 74% mortality, respectively, after ten weeks of inoculation. This mortality during the first week was 66%, 22% and 14% respectively. b) As a result of experimental pathogenicity, the symptoms produced were whorling, exophthalmia, ascites, haemorrhagic spots in the pelvic region, reddish protruded vent and body curvature. c) The LD50 calculated was 5.6x105 with 95% confidence limits of 1.37x105 to 2.35x106 cells/g body weight. 4. The primary and secondary humoral responses were ascertained against formalized Pseudomonas fluorescence injected intraperitoneally. a) The experimental fish administered with a dose of 4.8x1010 cells/g body weight on day 0 and day 12, gave antibody titer of 3.00+1.24, 2.66+0.27, 1.00+0.00 and 0.66+0.27, 2, 4, 6 and 8 weeks after the last injection, respectively. In an 8-week-old fish the control showed no antibodies after 2nd and 4th week but antibody titer of 0.33+0.27 was observed at the 6th and 8th week of the experiment. At a dose of 4.8x108 cells/g body weight an antibody titer of 2.00+0.81, 4.55+0.55, 1.33+0.54 and 1.33+0.72 was produced 2, 4, 6 and 8 weeks after the last injection, respectively. The 8-week-old fish at the above dose produced an antibody titer of 2.00+0.00 at 2nd week, 4.00+0.00 at 4th and 6th week whereas on the 8th week an antibody titer of 3.33+0.27 was noted. The group of fish injected with a final dose of 4.8x106 cells/g body weight produced in a 4-week-old fish an antibody titer of 3.00+0.00 on the 4th week, 2.33+0.54 on the 6th and 8th week and no antibodies were detected at the 2nd week of the last injection. The 8- week-old fish given the same dose produced no antibody titer at the 2nd week but a titer of 2.00+0.00, 3.66+0.27 and 3.50+0.55 were noted at the 4th, 6th and 8th week of the last injection of the antigen. b) The fish administered with a dose of 4.8x1010 cells/g body weight on day 0, day 12 and day 40, produced antibody titer of 3.60+0.27 on 2nd week and 4.00+0.47 on 4th and 6th week, whereas a titer of 3.50+0.55 was noted on the 8th week of the last injection. In the 8 week-old fish an antibody titer of 3.00+0.00, 4.00+0.47, 3.50+0.55 and 3.00+0.47 was observed 2, 4, 6 and 8 weeks after the last injection, respectively. At a dose of 4.8x108 cells/g body weight on day 0 and day 12 and a booster dose on day 40, in the 4-week-old fish, the antibody titers of 4.00+0.00,4.33+0.27,3.66+0.27 and 3.50+0,55 were observed 2, 4, 6 and 8 weeks after the last injection, respectively. In the 8 week old fish the above dose resulted in an antibody titer of 4.00+0.00, 3.00+0.47, 3.33+0.27 and 4.00+1.63 on 2nd, 4th, 6th and 8th week of the last injection. At a dose of 4.8x106 cells/g body weight administered to small fish on day 0, day 12 and day 40 produced antibody titer of 3.33+0.27 , at 2nd and 4th week, whereas a titer of 433+0.27 and 4.00+0.00 was observed at 6th and 8th week of the last injection. In the large fish when the same amount of antigen was injected, an antibody titer of 3.33+0.27 was obtained on 2nd week, while a titer of 4.22+0.27 was noted on the 4th and 6th week. A titer of 4.00+1.27 was observed on the 8th week after the last injection. 4. a) In the present study it has been observed that the administration of formalized P. fluorescence intraperitoneally produced proliferative response both in kidney and spleen. When different concentrations of the antigen were used to evidence immunological response, there was not much difference in the primary and secondary proliferative response of the kidney. The response in the primary treatment was, however, high during initial stages, whereas, the secondary response was higher throughout the experimental period b) It was also observed that the size of the spleen patches was increased in the fish during the secondary proliferative response than the primary proliferative response.
