Abstract
The research work is aimed at the isolation, structure elucidation and pharmacological evaluation of the chemical constituents of Sarcococca saligna. The completion of the doctoral project resulted in isolation and structure elucidation of thirteen new pregnane-type steroidal alkaloids, These steroidal bases were isolated by various simple as well as advanced chromatographic techniques. The structures of the compounds were established through detailed physical studies in conjunctions with the mass spectrometry (EIMS. HREIMS, FDMS and FABMS), basic spectroscopic techniques (infrared, ultraviolet,1 H-NMR and 13C-NMR) and highly sophisticated spectroscopic techniques such as COSY 45°, HOHAHA, NOESY, ROESY, HMQC, HMBC. ,Six known pregnane-type steroidal bases two of them first time from this plant were also isolated and identified. All the new' and known bases were subjected to various pharmacological screenings where some of them showed moderate levels of antibacterial, cytotoxic, phytotoxic and a-chymotrypsin inhibition activities. The new alkaloids isolated were named as sal1gnarine (79), salignarine-A(80), salignarine-B(81), salignarine-C(82), salignarine-D (83), salignarine- E(84), sarconidine (85). Kashmiramine (86), salignamine (87), Na-demethylsaracodine (88),sarcorine (89), E-salignone (90) and Z-salignone (91).Steroidal bases N-acetylirhidiamine-F (92) and epipachysandrine-A (36) were isolated for the first time from S. saligna. Steroidal bases alkaloid-A (39), alkaloid-C (40), saracodine (41) and saracodine (42) were also isolated and identified.
