Abstract
Effect of malathion, solfac and DDT was observed on the activity of enzymes (LDH, Cholinesterase, GOT, GPT) colorimetrically. HPLC and GLC (for estimation of pesticides) against the late 3rd instar larvae of Culex fatigans K.U. strain, G.I. strain and Ly. strain was done.

The LC50 value of malathion was found to be 0.225 ppm for K.U. strain, 0.44 ppm for G.I. strain and 0.5 ppm for Ly. strain. In the case of solfac the LC50 was 0.000165 ppm for K.U. strain, 0.00018 ppm for G.I. strain and 0.00045 ppm for Ly. strain, while the LC50 of DDT was 0.175 ppm for K.U. strain, 0.325 ppm for G.I. strain and 0.425 ppm for Ly. strain.

Biochemical estimation (colorimetry) revealed that malathion decreased 30% activity of LDH in K.U. strain, 58.33% in G.I. strain and increased 22.22% activity in Ly. strain after one minute incubation. Similar results were found after two and three minutes incubation.

Solfac decreased 33.33% LDH activity in K.U. strain, 50% in G.I. strain and increased 22.22% activity in Ly. strain after one minute incubation. Similar pattern of results were observed after two and three minutes incubation.

DDT decreased 50% LDH activity in K.U. strain, 66.66% in G.l. strain and increased 41.66% LDH activity in Ly. strain after one minute incubation. Similar results were. observed after two and three minutes incubation.

Malathion decreased 67.89% cholinesterase activity in K.U. strain, 19.96% in G.I. strain and increased 26.6% cholinesterase activity in Ly. strain after sixty seconds incubation. Similar results were found after ninety seconds.

Solfac decreased 66.661. cholinesterase activity in K.U. strain, 26.61. activity in G.I. strain and increased 641. cholinesterase activity in Ly. strain after sixty seconds incubation. Similar pattern of results were found after ninety seconds incubation.

DDT decreased 55.651. cholinesterase activity in K.U. strain, 41.741. in G.I. strain and increased 70.04% activity in Ly. strain, after sixty seconds incubation. Similar pattern of results were found after ninety seconds incubation.

In case of GOT treatment with malathion, decreased 41.17% activity in K.U. strain, 507. in G.l. strain and increased 35.13% activity in Ly. strain.

Solfac decreased 34.14% GOT activity in K.U. strain, 9% activity in G.I. strain and increased 75% activity in Ly. strain

DDT decreased 30% GOT activity in K.U. strain, 47.36% activity in G.I. strain and increased 53.33% activity in Ly. strain.

In case of GPT, treatment with malathion, decreased 6.97% activity in K.U. strain, 10.81% in G.I. strain and increased 6.06% activity in Ly. strain.

Solfac decreased 10.301. GPT activity in K.U. strain, 6.591. activity in G.I. strain and increased 12.96% activity in Ly. strain.

DDT decreased 26.31% GPT activity in K.U. strain, 43.921. activity in G.I. strain and increased 19.58% GPT activity in Ly. strain.

In case of Polyacrylamide gel electrophoresis (PAGE). malathion inhibit three bands of protein in K.U. strain, two bands in G.I. strain and increased intensity of three bands in Ly. strain as compared to control.

Solfac inhibit two bands in K.U. strain, one band in G.I. strain and increased intensity of four bands in Ly. strain as compared to control.

DDT inhibit three protein bands in K.U. strain, three bands in G.I. strain and increased intensity of three bands in Ly. strain as compared to control.

Acid phosphatase and alkaline phosphatase, bands also showed similar response to pesticides.

Estimation of malathion was done by HPLC. In the treated K.U. strain 800 ng, in G.I. strain 500 ng and in Ly. strain 300 ng malathion residue was recorded. The DDT treatment showed 125 ng in K.U. strain, 35 ng in G.I. strain and 30 ng in Ly. strain residues was recovered by HPLC.

Estimation of DDT by GLC was done in treated strains. In K.U strain 59.68 ng, in G.I. strain 18.3 ng and in Ly strain 4.33 ng residue was found.

Estimation of DDE by GLC in K.U. strain showed 11.6% ODE in K.U. strain, 25.2% in G.I. strain and 43.7% in Ly. strain, indicating resistance in Ly. strain with higher DDT dehydrochlorinase activity.
