Abstract
Experiments were carried out to study the toxicity and penetration effect of phosphice fumigant on Tribolium castancum Herbst., under bio-ecological conditions of Pakistan. Penetration effect of phosphine in grain was determined in special plastic tubes of 4, 8, 12, 20 and 28 feet length at temperatures 10, 15, 20, 25 and 30ÂșC. Complete mortality of adult beetles were obtained at phosphine concentration ranging from 0.950 to 0.380 for 4 feet, 1.200 to 0.615 for 8 feet 1.420 to 0.850 for 12 feet, 1.645 to 0.980 for 20 feet and 1.870 to 1.140 g/2.8m3 for 28 feet respectively

Experiments on large scale fumigation trials were also conducted in concrete godowns, warehouses and large sealed stores in order to determine LC50 and LC90 values of phosphine and methylbormide against eggs, larvae, pupae and adults of T. castaneum, at various temperatures, RH and exposure periods. Regression analysis indicates that the toxicity of periods. Regression analysis indicates that the toxicity of phosphine and methylbromide vary considerably according to the different stages of insects. The results showed 33% reduction in the dose of phosphine and 1.9 mg/1 of methylbromide under bio-ecological conditions prevailing in Pakistan. Significant inhibition of protein and enzymes (acid phosphatase, alkaline phosphatase, cholinesterase, glutamic oxaloacetic transaminase, glutamic-phruvic transminase, lactate dehydrogenase) was found. Inhibition of protein, at 30 g/1000 ft3 phosphine concentration was recorded as 66.65% and alkaline and acid comparatively less inhibition was found (20.27%) in LDH at 10 g/1000 ft3. Comparison of chromatogram was also done by TLC. The intensity of protein metabolites in control samples was found from High to poor whereas in treated insects at 10, 20, and 30 g/1000 ft3 (Phosphine) was found from Low to very poor. Similar response was found by histochemical localization and Gel scanning of different intensities in control and treated homogenate at 10, 15, 20 and 30 g/1000 ft3 (Phosphine). In all the control samples band A and D was found with high concentration of protein and rest ranging from Low to poor concentration, whereas in treated samples band A with Low-poor to very poor intensity was found and D with high performance liquid chromatography also revealed 5 protein peaks. In control samples proteins of peak, 2, 4, 5 was found in sufficient quantity while peak 3 was small and 6 was moderate

In treated extracts inhibition of some proteins and addition of some new protein/polypeptides was also observed. Slight variation in retention time was also recorded. Peak 3 was totally absent while tow new peaks P7 and P8 were observed
