Abstract

The present investigation was carried out on vinegar fly Drosophila melanogaster covering various toxicological aspects i.e. acute toxic effects of pesticides, effects on fecundity teratomorphic effects, biochemical effects and quantitative recovery of pesticides by HPLC. The under test compounds were RB-b (alone), RB-b with synergist, dimilin and danitol. For all the experiments conducted, feeding method was employed. The LD50 of RB-b (alone), RB-b (synergized), dimilin and danitol were calculated as 2, 1.6, 0.0312 and 0.001 μg/larva, respectively. Efficacy of tested compounds was as danitol > dimilin > RB-b (synergized) > RB-b (alone) with reference to toxicity, fecundity and teratomorphic effects. Variations were found after comparison of all respective compounds Biochemical analysis was done on total protein content, total esterase, acetylcholinesterase, Phosphomonoesterrases (acid and alkaline phosphatrase), transaminases (GOT and GPT) and nucleic acids (RNA and DNA). On the basis of biochemical analysis versus the tested compounds did not follow a regular order of inhibition Among respective compounds inhibition observed by RB-b (alone) was 55.8% (total protein), 39.8% (total esterase), 45.1% (acetylcholinesterase), 47.5% (acid phosphatase), 49.5% (alkaline phosphatase), 82.1% (GOT), 47.4% (GPT), 74.33% (DNA) and 31.83% (RNA). So the maximum inhibition of enzyme GOT was by the treatment of RB-b (alone) RB-b (synergized) inhibited total protein (51.9%), total esterase (42.4%), acetylcholinesterase (45.1%), acid phosphatase (50.3%), alkaline phosphatase (52.6%), GOT (69.3%), GPT (52.6%), DNA (84.4%) and RNA (53.3%). The maximum inhibition of DNA contents occurred by RB-b (synergized) Dimiln caused inhibition in total protein (51.2%), total esterase (40.7%), acetylocholinesterase (61.6%), acid phosphatase (61.4%), alkaline phosphatase (47.8%), GOT (68.8%), GPT (51.2%), DNA (81.3%), contents decreased upto 53.3%. The maximum inhibition occurred in the case of DNA by dimilin Whereas danitol (fenpropathrin) inhibited 55.7% (total protein contents), 46.4% (total esterase), 61.6% (acetylcholinesterase), 71% (acid phosphatase), 70.9% (alkaline phosphatase), 63.1% (GOT), 48.5% )GPT) 81.3% (DNA) and RNA contents decreased upto 45.2% HPLC method was used for the quantitative analysis in treated larvae after 24 hours of treatment. The recovery of dimilin and danitol was 0.60% μg/10 μl and 0.43 μg/μl respectively. Recovery of dimilin was comparatively higher in treated insects than danitol it means that dimilin has low toxic effect
