Abstract
Present thesis is divided into two parts. Part-A involves screening of one hundred and sixty five plant materials obtained from one hundred and twelve important indigenous medicinal herbs of Pakistan. Part-II records the investigations of alkaloidal constituents of Sphaeranthus indicus Linn

Part-I. For screening of medicinal plants collection was made from different parts of the country the extracts were obtained from different parts of plants using 50% ethanol. Antibacterial activity was determined against eighteen different gram negative and gram positive species of bacteria involved in common infections. Twenty Five percent plant extracts exhibited broad spectrum activity against all the organisms used in the study. Thirty three percent showed narrow spectrum activity against either Gram negative or Gram positive bacteria. Thus a total of approximately fifty eight percent of extracts exhibited antibacterial activity.

Among one hundred and sixty five plant materials screened flowers of Sphaeranthus indicus showed most promising activity. The crude ethanolic extract of this plant was fractionated into alkaloidal and non-alkaloidal fractions by chemical method, and antibacterial activity. From the alkaloidal fraction four previously unknown alkaloids Sphaeranthine I (s1), Sphaeranthine IV (S4), Sphaeranthine VI (S6) and Sphaeranthine VII (S7) were obtained by chromatographic method. Alkaloids S1 exhibited antibacterial activity

Bioautographic studies were also carried out to detect the active principal on TLC plate. The results of bio-autography were in conformity with the results obtained by cylinder plate method. Comparison of the crude extract of Sphaeranthus indicus with the existing commercial antibiotics was also carried out and in all cases the plant extract was found to be much more potent than the antibiotics used in the study

Part-II From the flowers of Sphaeranthus indicus has been isolated four new alkaloids named as Shpaeranthine I (S1), Sphaeranthine IV (S4), Sphaeranthine VI (S6) and Spheranthine VII (S7) investigations. The mass spectrum of S1 and S4 showed the molecular ion peak at m/e 268, m/e 273 indicating the molecular formula as C12H8N2O5 and C14H8O4 respectively. Whereas the mass spectrum of S6 and S7 were recorded at m/e 266 and m/e 254 exhibiting the molecular formula as C12H8N2O5 and C14H8O4 and C17H18N2O respectively. IR, UV, NMR and C-13 NMR of these compounds revealed some chemical and spectral features
