Present research work was undertaken to investigate the root-knot formation in banana plant in University Campus, Karachi. The nematode was identified as meloidogyne javani- ca. Pathogenicity of this nematode to banana root was also studied in detail. The information on pathogenicity will help in controlling this parasite in banana and other plants and will in further research on this topic. As this problem is of great concern to the economy of the country. For this problem is of great concern to the economy of the country. For this purpose thick sections cut on a glass slide with sharp blade were prepared and mounted permanently in Canada balsam by using stsandard techniques which are described in material and methods

Scanning electron microscopy was also carried out to confirm the identification. For this purpose thick sections of infected banana roots were fixed in 4% gluteraldehyde for 24 hours, dehydrated with amyl acetate, after critical point drying mounted on stubs, coated with gold and photographs were taken on a Cambridge S 180 steroscan through the courtesy of EM. The present specimens of M. Javanica characterized by possessing a papilla like structure at the lower margin of vulva. Lateral ridges reaching to tail terminus, rounded dorsal and ventral arches, striae bending towards vulval region. The ultrastreucture of head region confirms that the present specimens are similar to M. javanica as the medial lips and labial disc are fused together and form one smooth chitinous structure, almost as wide as head region in lateral view. The head is not distinctly set off from rest of the body

Gross morphology and productivity of banana plant was affected due to this infection. Yellowing of leaves, stunting reduced growth of plant and less fruit production was observed. Roots were galled, galls were either found individually or in clusters

Study of the sections of infected banana root with light and scanning electron microscopy revealed that severe damage was caused to the root tissue. Giant cell formation at the site of infection was a common finding. Cells along the pathway of migration of nematode were damaged, and canal leading to cavity was obvious. Cavity formation was observed in banana root due to this infection. Females were found inside the cavity close to which giant cells and damaged cells were observed. Xylem vessels were severely disturbed, surrounding cells become oblong, with outline of vessel cavity slightly irregular and abnormal structures occupying the vessel cavity
