Although introduction of proton-pump inhibitors to the classic anti-ulcer therapy had revolutionized treatment of the peptic ulcers and other gastrointestinal disorders but there was still no complete cure of the disease. Generally, there were relapses and ineffectiveness of different drug regimens, production of adverse and side effects, emergence of resistance to drugs and higher costs of these therapeutic agents. Therefore, present studies were designed to find out some possible radical or ideal anti-ulcer drugs from the indigenous medicinal plants. Six reputed and frequently used folkloric medicinal plants were selected for evaluation of their anti-ulcer activities in albino rats. Efficacies of the powdered plants and their extracts in water and ethanol were determined in indomethacin-induced ulcerated rats. On the basis of the data obtained, Nigella sativa seeds and Phyllanthus emblica fruits were selected for further activity-oriented fractionation studies as they had showed most promising anti-ulcer properties. The ethanolic extract of N. sativa seeds and aqueous extract of P. emblica fruits were fractionated into ethyl acetate, chloroform and butanol solvents. On the basis of anti-ulcer potentials, ethyl acetate fractions were subjected to column chromatography. The purified fractions of both N. sativa seeds and P. emblica fruits were tested for their anti-ulcerogenic activities in stress-induced, indomethacin- and histamine-induced gastric ulcers in the albino rats. Various parameters including gastric juice volume, pH, acid-output, ulcer index, mucus secretion and peptic activity showed that NS-EA 51 and PE-AqA 47 were the most potent anti -ulcerogenic fractions and gave results comparable to famotidine. In addition, the anti-Helicobacter pylori activities of NS-EA 51 and PE-AqA 47 fractions were also found comparable to amoxicillin and clarithromycin. The present studies also suggested that NS-EA 51 might have exerted its antiulcer activity due to its proton pump (H+/K+-ATPase) inhibitory property as revealed by the fluorescence quenching of acridine orange dye. On the other hand, PE-AqA 47 fraction did not interfere with H+ /K+ -ATPase and has possibly showed antiulcerogenic effect due to its anti-oxidant property. Finally, the acute toxicity and behavioral pattern studies in rats showed that both NS-EA 51 and PE-AqA 47 fractions are quite safe and may ultimately prove to be ideal antiulcer remedies.
